Some aryl-oxazolines have been described as active substances on the central nervous system with diverse effects and applications as depressants, anesthetics, anticonvulsants, etc. In the present paper, the primary neuropharmacological characterization of 2-phenyl-4, 4-bis-(hydroxymethyl)-2-oxazoline, obtained by a method of synthesis under microwaves irradiation as an energy source, was made. The application of Irwin's test showed that this compound has a depressive effect on the central nervous system, with a marked inhibition of the motor activity and muscular tone and with loss of postural reflexes. At moderate doses, these effects were accompanied with signs of central excitation. These characteristics suggested an action of peripheral type, confirmed by the blocking effect of the contorsions in the peritoneal irritation test caused by acetic acid 0.8 % and the reduction of the potential amplitude of the composed neuromuscular action during the supramaximal stimulation of the cyatic nerve, tests that evidenced a local anesthetic action for 2-phenyl-4, 4-bis-(hydroxymethyl)-2-oxazoline.
Algunas aril-oxazolinas han sido descritas como sustancias activas sobre el sistema nervioso central, con efectos y aplicaciones diversas como depresoras, anestésicos, anticonvulsivantes, etc. En el presente trabajo se realizó la caracterización neurofarmacológica primaria de la 2-fenil-4,4-bis-(hidroximetil)-2-oxazolina, obtenida por un método de síntesis bajo radiación de microondas como fuente energética. La realización de la prueba de Irwin mostró que este compuesto presenta un efecto depresor sobre el sistema nervioso central, con una notable inhibición de la actividad motora y del tono muscular, con pérdida de los reflejos posturales. A dosis moderadas estos efectos se acompañaron de signos de excitación central. Estas características sugirieron una acción de tipo periférica, confirmado por el efecto bloqueador de las contorsiones en la prueba de irritación peritoneal por ácido acético al 0,8 % y la disminución de la amplitud del potencial de acción neuromuscular compuesto durante la estimulación supramáxima del nervio ciático, pruebas que evidenciaron una acción anestésica local para la 2-fenil-4,4-bis-(hidroximetil)-2-oxazolina.
